Early events in pluripotency exit

Embryonic stem cells needs to exit pluripotency to be able to initiate differentiation programs. Resolution of pluripotency involves many steps including rewiring of chromatin and genetic networks, enhancer de-/re-commissioning, and altered cellular signaling. But, what are the first events that induce these global changes? When stemness factors are removed, ES cells commit to exit pluripotency and initiate differentiation within the first 12 hours. By 24 hours, differentiation is well underway, despite persistence of some epigenetic marks and pluripotency factors.

We focus on the first few hours of pluripotency exit to identify factors that govern and execute the early events in resolution of stemness.
